題:Skin Pigmentation Pathways and Melanoma Risk

旨:Melanoma risk is well known to be strongly linked to UV exposure. Yet despite
this important linkage, melanoma incidence increases relentlessly, at one of the
steepest rates of all human cancers. Prevention approaches that include education,
sunscreen, avoidance of tanning beds, and sun-safe behavior messages are excellent in
principal, but have not as yet produced sufficient decreases in melanoma incidence.
Here we present studies which explore cutaneous responses to ultraviolet radiation
exposure. These results indicate the presence of multiple cutaneous responses and
pathways that are triggered by UV and which modulate not only skin signaling, but also
systemic effects of this cutaneous UV exposure. These pathways include pigmentation
or tanning. Other studies will describe a murine “redhair/lightskin” melanoma model
of melanoma which identifies roles for pigmentation intermediates in melanoma formation.
Finally, the MITF transcription factor plays central roles in both melanocyte and
melanoma growth, survival, and differentiation. Studies will be described which
characterize several MITF transcriptional targets, some of which may represent novel
therapeutic opportunities.
