●活性の定義
活性化サケ精子 DNA を鋳型／プライマーとして用い、下記の活性測
反応液中にて 74℃において、30 分間に 10 nmol の全ヌクレオチドを酸
溶性沈殿物に取り込む活性を 1 U とする。
●活性測定用反応液組成
25 mM TAPS 緩衝液 (pH9.3, 25℃ ), 50 mM KCl,
2 mM MgCl2 , 1 mM 2- メルカプトエタノール ,
各 200 μ M dATP・dGTP・dTTP, 100 μ M
〔α -32P〕dCTP, 0.25 mg/ml 活性化サケ精子 DNA
●純度
・10 U の本酵素と 0.6 μ g のλ -Hin d III 分解物とを 74℃、１時間反応さ
せても DNA の電気泳動パターンに変化は起こらない。
・10 U の本酵素と 0.6 μ g の supercoiled pBR322 DNA とを 74℃、１時
間反応させても DNA の電気泳動パターンに変化は起こらない。
・10 U の本酵素と 0.6 μ g のλ DNA とを 74℃、１時間反応させても
DNA の電気泳動パターンに変化は起こらない。
●用途
・Polymerase Chain Reaction (PCR) 法による DNA 増幅
・DNA シーケンシング
● PCR 検定
・λ - DNA を用いた PCR 反応（増幅産物 8 kb）において良好な増幅が
見られることを確認している。
・ヒト Genome DNA を鋳型とした single copy gene の PCR 反応 ( 増幅産
物約 3 kbp ) において良好な増幅が見られることを確認している。

● Cool Start 法
PCR の際の非特異的な増幅を抑える手法としては、ワックスや、専
用試薬、酵素を用いた Hot Start 法が知られていますが、特別な酵素
や試薬を必要としない Cool Start 法により簡便に PCR 時の非特異的
増幅を抑えることができます。
＜プロトコール＞
(1) 試薬をすべて氷上に置いておきます。
(2) 試薬分注後の反応チューブは、即氷上に置きます。（チューブに加
える試薬の順番は問題になりません。調製後 30 分たってから反応し
ても大丈夫です。）
(3) サーマルサイクラーはスタートするだけの状態にしておきます。
（設定は既存のプログラムで OK です。）サンプル数が多い場合は、サ
ンプルプロッグを 4℃にあらかじめセットしておいてください。
(4) 反応チューブをサーマルサイクラーにセットし、即スタートさせま
す。
