バイオイメージングセンターセミナー 濱崎 万穂 先生 (大阪大学大学院 医学系研究科 遺伝医学講座・准教授/JSTさきがけ) 哺乳動物細胞における オートファゴソーム形成初期過程の解析 日時:3月12日 木曜日14時∼ 場所:柏キャンパス生命棟地下1階講義室 要旨: Autophagy is a tightly regulated intracellular degradation system that plays fundamental roles in cellular homeostasis. Autophagy is induced by starvation, bacterial infection, accumulation of aggregate proteins, organelle damages and so on. Autophagy is initiated by nucleation of flattened vesicles termed isolation membranes that get elongated to engulf its target and become autophagosomes. Autophagosomes then fuse with lysosomes to degrade the contents. Autophagosomes are quite unique compared to other organelles as they form only when they are required from the scratch. The site of this organelle formation and the origin of their membranes have been a key question to understand the molecular mechanism of autophagosome formation. In this study, we examined the initial stage of autophagosome formation site. To determine the autophagosome formation site, we used Atg14L (a component of initial autophagosome formation machinery) and Atg5 (an autophagosome marker covers from the beginning) under starved condition. We observed the ER-Mitochondria contact sites play an important role for the autophagosome formation. This provides new insights into the autophagosome formation.
