Description :
This DNA is a vector which is available to enforcement cloning system.
This DNA contains chloramphenicol reistant gene and lethal gene rpsL
(streptomycin-sensitive ribosomal protein) at downstream region of
tryptophan promoter. Since this is designed for multi-cloning site possessing amber mutant and 39 restriction sites on rpsL gene, normal rpsL will
not be expressed if a target gene is inserted into those restriction sites.
Therefore, only targeted clones can be grown, because uninserted clones
can not be grown in trpR －, rpsL － and supE － host like E. coli TH2.

本製品は、クロラムフェニコール耐性遺伝子とトリプトファンプロモー
ターの下流に致死遺伝子 rpsL （ストレプトマイシン感受性リポゾームタ
ンパク質）をもつ、Enforcement Cloning Vector である。rpsL 遺伝子上
にはアンバー変異と 39 種類の制限酵素切断部位を持つマルチクローニン
グサイトが設計されており、それらの部位に外来遺伝子が挿入されると
正常な rpsL が発現されない。そのため、E. coli TH2（trpR624、rpsL20、
supE44 ）を用いることにより、外来遺伝子が挿入されていないクローン
はストレプトマイシンを含む培地では生育阻害が生じ、目的の外来遺伝
子を持つクローンのみが生育し、容易にクローンの選別が可能となる。
