Recent advances in the sensitivity of next generation sequencing (NGS)
and gene expression technologies have enabled the analysis of complex
cell micro environments. However, there remains the challenge to
separate and identity homogeneous cell populations prior to analysis.
Here we describe how the power of flow cytometry can be exploited to
isolate, characterize, and purify single cell populations from complex cell
populations, prior to gene expression profiling. We describe a complete
workflow starting with heterogeneous cancer cells that can be isolated at
rapid speed by single cell depositions into 96 well plates using the newly
developed flow cytometer from BD Biosciences, the BD FACSJazz™. We
demonstrate the use of new analytical tools such as index cell sorting
that can help track those individual cells during and after sorting. Finally
we demonstrate that the single cell populations are of high fidelity and
suitable for gene expression analysis using the NanoString nCounter®
Analysis System with gene expression assay panels capable of
simultaneously measuring up to 800 gene expression targets.
