2 Fakultas Kedokteran Gigi Departemen Periodonsia Tahun 2014 Hazwani Izyan Bt Azahar Efektivitas Ekstrak Teh Hijau (Camellia sinensis) terhadap Pertumbuhan Bakteri Porphyromonas gingivalis secara In Vitro x + 31 halaman Penyakit periodontal dikaitkan dengan keberadaan bakteri negatif Gramm yang menyebabkan kehilangan perlekatan jaringan periodontal tulang alveolar yang mendukung gigi. Porphyromonas gingivalis adalah salah satu patogen utama pada penyakit periodontal dan berada pada permukaan biofilm subgingiva dan berkolonisasi di mukosa mulut. Teh hijau mengandung kadar polifenol yang tinggi terutama catechin dan bersifat antioksidan, antikarsinogenik dan bersifat hipolipidemik. Tujuan dari penelitian ini adalah untuk melihat efektivitas ekstrak teh hijau terhadap bakteri Porphyromonas gingivalis dengan memperhatikan nilai Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM). Daun teh hijau sebanyak ±400 gram dikeringkan dan dihaluskan, kemudian diekstraksi dengan pelarut etanol 70% sehingga diperoleh ±45 gram ekstrak kental teh hijau. Penentuan KHM dilakukan dengan metode dilusi yaitu ekstrak kental teh hijau disuspensikan dengan Tryptic Soy Broth (TSB) dan dilakukan pengenceran ganda sehingga diperoleh konsentrasi 100%, 50%, 25%, 12,5% dan 6,25% yang masing-masing terdiri dari 5 sampel. Masing-masing konsentrasi tersebut diambil sebanyak 1 ml, ditambahkan 1ml suspensi bakteri, divorteks dan diinkubasi 37 oC selama 24 jam pada inkubator CO2. Kekeruhan diamati dan dibandingkan dengan kontrol Mc Farland untuk menentukan KHM. Kemudian tiap kelompok dicampur menggunakan vorteks dan diambil 50 l, diteteskan ke Tryptic Soy Agar (TSA), direplikasi 5 petri, didiamkan 15-20 menit lalu diinkubasi 37 oC selama 24 jam pada inkubator CO2. Perhitungan jumlah koloni bakteri dilakukan dengan metode Drop Plate Mills Mesra untuk menentukan KBM. 3 Hasil penelitian menunjukkan bahwa ekstrak teh hijau mempunyai efek antibakteri terhadap Porphyromonas gingivalis pada konsentrasi terendah, 6,25% masih dapat membunuh bakteri dengan hasil perhitungan koloni 0 CFU/ml. Porphyromonaas gingivalis is one of the main pathogen that leads to periodontal disease and located on biofilm of the subgingiva surface and colonized in the mucosa of the mouth. Green tea contains polyphenol substance especially catechin which act as antioxidant, anticarcinogenic and hypolipidemic. The purpose of this study is to observe the effectiveness of the green tea extract towards bacteria Porphyromonas gingivalis by using Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Green tea leaves was hand picked ±400 grams, dried and was blendered until in the form of powder, later the extraction was taken using percolation method using ethanol 70% as solvent until ±45 grams of thick green tea extract was formed. Minimum Inhibitory Concentration (MIC) was obtained using dilution method which green tea extract was suspended with Tryptic Soy Broth (TSB) and double dilution method was used until 5 samples of each concentration between 100%, 50%, 25%, 12,5% and 6,25% was formed. 1 ml from each concentration was taken, 1 ml bacteria suspension was added, mixed and incubated 37 oC for 24 hours in CO2 incubator. The cloudiness of each samples was observed and compared with Mc Farland control for determination of Minimum Inhibitory Concentration (MIC). Later on every concentration from previous test tube was mixed using vortex and 50 l was taken, and been dropped into Tryptic Soy Agar (TSA), 5 petries replication was formed and let it set for about 15-20 minutes and incubated 37 oC for 24 hours in CO2 incubator. The 5 calculation of the total bacteria colony using Drop Plate Mills Mesra method for Minimum Bactericidal Concentration (MBC). The results shown that green tea extract has anti bacterial effect on Porphyromonas gingivalis and eventhough in the lowest concentration which is 6,25%, the availability to kill bacteria with total count 0 CFU/ml.
