High Titer AAV Preparation without a Helper Virus
• Simply cotransfect HEK 293T cells with three plasmids (pAAV
with gene of interest, pRC2-mi342, and pHelper) to generate
recombinant AAV particles.
• Produce AAV particles at very high titer—expression of human
microRNA-342 in this system doubles titer.
Easy and Efficient AAV Particle Extraction
• Can be used for any AAV serotype.
• Increases viral particle recovery by at least 3-fold as compared
to conventional freeze-thaw methods.
• Resulting viral particle solution contains low amounts of host
DNA and protein contamination.
Simple and Fast Purification of AAV Particles
without Ultracentrifugation
• Column-based purification yields high yields of highly pure
AAV particles in ~4 hours.

or ELISA methods.
