In this talk I will first highlight our efforts at an exciting frontier in molecular pharmacology--the identification of a functionally relevant protein target of general anesthetics. This work built on our discovery of the first fluorescent general anesthetic, 1-aminoanthracene, and subsequent Harvesting RNA with light. I will then highlight our efforts to advance the field of transcriptomics by making it possible to elucidate how RNA populations vary between different cells in living brain tissue. This work builds on my lab’s long-standing interest in designing light-activated oligonucleotides to manipulate gene expression with high spatiotemporal resolution. For capturing mRNA, we developed Transcriptome In Vivo Analysis (TIVA) technology, which also employs light microscopy to control the function of caged oligonucleotides.
